چکیده
Two sisters with a previously unreported megaloblastic anemia unassociated with a deficiency of either folic acid or vitamin B12 are described. Deficiencies of these vitamins were ruled out by standard studies. All other previously reported forms of megaloblastic anemia not secondary to a vitamin deficiency, such as orotic aciduria, were also excluded by appropriate studies. Optimal hemoglobin responses were obtained after the administration of large amounts of both vitamin B12 and folic acid. Because of this hemoglobin response, the conversion of deoxyuridine-5-monophosphate to deoxythymidine-5-monophosphate in vitro was examined in bone marrow samples from both patients using a modification of a method described by Killmann.18 This preliminary step in DNA synthesis was found to be normal. The results of this test and the optimal hemoglobin response after administration of both vitamins suggest that both folic acid and vitamin B10 may be necessary at some other preliminary step in DNA

متن کاملMegaloblastic Anemia: A Drug-Induced Disorder
Megaloblastic anemia is categorized by distinguished hematopoietic cell morphology, unproductive hematopoiesis, induced by folic acid and/or vitamin B12 deficiency [1]. Ineffective DNA synthesis in hematopoietic progenitor cells or myelodysplastic syndrome leading to congenital disorders is commonly responsible for the megaloblastic anemia. In western populations, cobalamin (vitamin B12) defici...

Megaloblastic erythropoiesis is known to occur in patients with chronic hemolytic anaemia. It has been described in association with sickle cell anemia (Oliner and Heller 1959 ), thalassaemia (Robinson and Watson 1963 ), congenital spherocytosis ( Delamore et al. 1961), and acquired hemolytic anemia ( Forshaw and Harwood 1963). In a survey of the literature up to 1962, Robinson and Watson (1963...
متن کاملCongenital Methylmalonic Aciduria - Homocystinuria With Megaloblastic Anemia : Observations on Response to Hydroxocobalamin and on the Effect of Homocysteine and Methionine on the Deoxynridine Suppression Test
Congenital methylmalonic aciduria-homocystinuria, a disorder with an incompletely defined cobalamin abnormality. has not been accompanied by megaloblastosis in most of the initially described cases. This has raised questions about the exact role of cobalamin in relation to megaloblastic anemia. Therefore, we present our observations on a patient with this syndrome whose manifestations conformed...
